Eind 15e eeuw was Leiden de grootste stad van het graafschap Holland. De welvaart was vooral te danken aan de lakenindustrie. In de 16e eeuw kwam daar een einde aan toen een hevige strijd werd gevoerd met de Spanjaarden tijdens de Tachtigjarige Oorlog. Voor Leiden eindigde deze strijd met een overwinning en het Ontzet van Leiden wordt tot op de dag vandaag jaarlijks groots gevierd. Immigranten uit de Zuidelijke Nederlanden gaven de wolnijverheid een nieuwe impuls en de stad werd meerdere malen uitgebreid. De eerste universiteit van de Noordelijke Nederlanden werd in Leiden opgericht en tot op de dag van vandaag is Leiden een bruisende studentenstad. De historische route voert door het hart van de stad, langs imposante kerken en monumenten en verborgen steegjes en hofjes.
